Cancer is an outcome of low immunity level of people,change in diet, sedentary life style,stress, pollution, Drinks, medicine etc.

to imagine the basic cayuse of cancer or for that matter any serious problems in anyfield of research or inventions

How will your solution make a difference

The difference is in my mind set- If I want to improve a water pump I become water and pass through the pump and I get the results automatically, ie.become one with the subject giving proven results eg Apple for diabetes,etc docts attached.

What is your proposed solution

I first need to dig out facts on various points as per my thoughts and then apply my imagination based on the data collected and fix up the solution in my mind itself ,and work only to prove how my thinking is correct . I normally work on this mind set and have given proven results, study my inventions and results and then you can relaise. This happens because I am a passionate inventor and it is my liking to invent without anyexpectations

Immunotherapy responders vs nonresponders

Why do some cancer patients receiving checkpoint blockade or other immunotherapy respond and others not? How can we study this? Is deep sequencing of the cancer the answer?

How will your solution make a difference

If we could understand why some patients respond well to immunotherapy we can help those that do not. This under standing can be applied to cancer vaccines, CAR-T cell, and checkpoint blockade immunotherapies.

What is your proposed solution

Sequencing the neo-antigens of the tumors before treatment, during and after conclusion of treatment would allow scientist to study how the MHC I expression of the cancer changes as a result of immunotherapy. Microarray studies of such patients also would give data on how the tumor changes during the course of immunotherapy. Establishing bio-specimen tissue banks will allow us to continue to ask new questions as our understanding grows.

This would not be a cure for cancer but it could make a huge difference in the way patients feel when going through the treatments for their cancer and for those patients who have no treatments available for them.

The combination of all 3 could possibly systemically eradicate cancer without causing other life-threatening damage to patients' bodies now seen from radiation, chemotherapy and surgery.

What is your proposed solution

One small study at Hopkins is looking at cryoablation+immunotherapy for prostate cancer now. Mayo clinic doing clinical trial looking at cryoablation of tumors in lungs now. Combo of all 3 could have potential to create essentially a personalized vaccine.

Patient-derived xenografts (from cancer tissue biopsies) can be grown in immunosupressed and humanized mice. CAR-T cells (expressed via a scFv library) can be screened in a library format, allowing for rapid discovery of new targets.

How will your solution make a difference

At present, CAR development is a slow process based on single antigenic targets. Here we will go "target blind" in a large screening assay for scFv targets and definitively determine entire families of CAR-effector T cell targets for cancer.

What is your proposed solution

Using immunosupressed mice (i.e. NSG bearing human marrow-derived hematopoietic cells), patient xenografts will be established. This system models the tumor microenvironmentl seen in patients. Both anti-stromal and anti-tumor CAR activity will be tested. Unique, will be the high-throughput screening of CAR-T cells using an entire scFv library, encoding a cDNA library level of binding diversity. Thus, unique specific scFv will be discovered.

Therapies that augment NK cell activation and prevent NK cell inactivation will provide complementary therapies to those that mobilize T cells. Because NK cells augment T cell responses, such approaches may also improve existing therapies.

What is your proposed solution

Better understanding is needed of the processes that activate and recruit NK cells to tumors, and inactivate them once there. More research is needed to probe the molecular bases of activation and inactivation. Understanding these processes will enable the design of strategies to augment NK cell activity and prevent NK cell inactivation in the tumor environment. Efforts to test methods to better target NK cells to tumors are also called for.

Low doses of radiotherapy, TLR ligands and checkpoint antibodies given to a single site of disease, thereby triggering a systemic anti tumor T cell immune response.
This avoids having to identify the tumor antigens and it avoids toxicity because of the low doses and localized delivery of the therapy.
This is practical, cost effective and potentially scalable worldwide.
The optimal combination of injectable agents remain to be determined.
